if you are going 3d with this does that number assume a straight line through the planet from point nemo to the nearest assumed land based humans or does it goes the long way around like if you were looking at a flat map?

Along the surface here, since we can't currently travel across the globe shorter or faster by tunnelling straight the earth. Even so, 400km to the ISS trumps the distance to the nearest land by a huge margin, regardless of whether OP means the surface or the 'chord' distance - straight through.

How close something is doesn't care about how fast you can get there. If I'm on one side of a 10 inch thick piece of metal and you're on the other side but the only way to get to the other side is to walk 30 feet around the long way that doesn't make me more than like 13 inches away from you.

I agree with guest. The diameter of earth is roughly 12,000 km, so I can’t imagine how cutting through at any point would do enough to significantly change this piece of trivia.

"Land" can also mean the seafloor itself.

I had never thought about it and I'm starting to get the feeling of pure loneliness one must get when they reach this point. It must be so absolute and beautiful, but depressing and terrifying.
