I'm fairly sure the point is that her ex thought only married or not was an important question to ask a woman. Her PhD might be in environmental science, or race relations, or palaeontology.

Yeah, I thought it was obvious the ex was trying to figure out if she was single or not? but I always read that not like he was trying to flirt with her but trying to gauge how successful she was in relationships, and her response is that, it doesn't matter what her relationship status is bc she is successful in life.

The joke is that the person who commented is stupid. The joke is not about her education or her martial status. And it IS a joke, by the way.
